I am currently reading a book by Allison Weir entitled " Mary, Queen of Scots and the Murder of Lord Darnley" The author promises an in depth look at the circumstances leading to Darnley's death & an unbiased conclusion as to whether Mary may have been complicit in the deed. As I'm only on pafe 107 of 586, I have no idea.
I can only conjecture probably not. However, if she had a clue, I don't think she would have rushed to intervene to save her husband.
